The localization of acyl coenzyme A-carnitine acyltransferases in rat liver cells.

چکیده
The distribution of acetyl coenzyme A-carnitine acetyltransferase (EC 2.3.1.7) and of palmityl-CoA-carnitine palmityltransferase (EC 2.3.1) in subcellular fractions of rat liver has been studied. The distribution of carnitine pahnityltransferase in the different fractions obtained with differential centrifugation and with centrifugation in a steep sucrose gradient correlated closely with the distribution of the mitochondrial marker enzymes, glutamate dehydrogenase (EC 1.4.1.3) and D-phydroxybutyrate dehydrogenase (EC 1.1.1.30). Carnitine acetyltransferase also most probably is a mitochondrial enzyme. The results obtained suggest that this enzyme is present in a mitochondrial population not identical with the population rich in carnitine palmityltransferase and in the mitochondrial marker enzymes used. Most of the carnitine acyltransferases were found in the membranous part of the mitochondria. The findings presented are consistent with the hypothesis that the carnitine acyltransferases are present in the membranous part of the mitochondria which is the barrier for the acyl-CoA molecules.

Carnitine acyltransferase and acyl-coenzyme A hydrolase activities in human liver. Quantitative analysis of their subcellular localization.
The subcellular localizations of carnitine acyltransferase and acyl-CoA hydrolase activities with different chain-length substrates were quantitatively evaluated in human liver by fractionation of total homogenates in metrizamide density gradients and by differential centrifugation. Peroxisomes were found to contain 8-37% of the liver acyltransferase activity, the relative amount depending on t...
متن کاملCrystal structure of rat carnitine palmitoyltransferase II (CPT-II).
Carnitine palmitoyltransferase II (CPT-II) has a crucial role in the beta-oxidation of long-chain fatty acids in mitochondria. We report here the crystal structure of rat CPT-II at 1.9A resolution. The overall structure shares strong similarity to those of short- and medium-chain carnitine acyltransferases, although detailed structural differences in the active site region have a significant im...
متن کاملImportance of experimental conditions in evaluating the malonyl-CoA sensitivity of liver carnitine acyltransferase. Studies with fed and starved rats.
The experiments reconfirm the powerful inhibitory effect of malonyl-CoA on carnitine acyltransferase I and fatty acid oxidation in rat liver mitochondria (Ki 1.5 microM). Sensitivity decreased with starvation (Ki after 18 h starvation 3.0 microM, and after 42 h 5.0 microM). Observations by Cook, Otto & Cornell [Biochem. J. (1980) 192, 955--958] and Ontko & Johns [Biochem. J. (1980) 192, 959--96...

Peroxisomal beta-oxidation of branched chain fatty acids in rat liver. Evidence that carnitine palmitoyltransferase I prevents transport of branched chain fatty acids into mitochondria.
Fatty acid beta-oxidation was investigated in highly purified mitochondrial and peroxisomal preparations from rat liver. Under isotonic conditions, pristanic and homophytanic acid beta-oxidation in purified peroxisomes was severalfold greater compared to the oxidation in purified mitochondria. Branched chain fatty acid beta-oxidation in purified mitochondria was very low, and the oxidation was ...
